چکیده

Recently, long noncoding RNAs (lncRNAs) have been reported to play important role in the pathogenesis of various cancers. However, functions RNF185-AS1 hepatocellular carcinoma (HCC) remain unknown. The expression HCC cells and tissues was measured by quantitative real-time polymerase chain reaction (qRT-PCR). effects on tumor cell proliferation, migration invasion were assessed Cell Counting Kit-8 (CCK8) assay, colony formation transwell assay. luciferase reporter RNA-binding protein immunoprecipitation qRT-PCR Western blot performed explore confirm interaction between miR-221-5p integrin β5. progression explored through vivo experiments. highly expressed lines. High levels correlated with advanced TNM stage, distant metastasis a poorer overall survival rate. knockdown inhibited invasion. Additionally, acted as sponge for β5 identified target gene miR-221-5p. Rescue assays showed that inhibitor or overexpression rescued function migration, Moreover, we found growth metastasis. Our study demonstrates is new oncogenic lncRNA suggests RNF185-AS1/miR-221-5p/integrin axis might be potential therapeutic HCC.

Long noncoding RNA NNT-AS1 promotes hepatocellular carcinoma progression and metastasis through miR-363/CDK6 axis

Long non-coding RNAs (lncRNAs) have been tested to act as important regulator in liver cancer genesis and progression. LncRNA Nicotinamide Nucleotide Transhydrogenase-antisense RNA1 (NNT-AS1) has been reported to participate in the tumorigenesis. However, the exact molecular mechanism of NNT-AS1 in hepatocellular carcinoma (HCC) is still unknown. In present study, our team identified the up-reg...

miR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting

Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...

EZH2 promotes hepatocellular carcinoma progression through modulating miR-22/galectin-9 axis

BACKGROUND Recent studies have shown that interferon-γ (IFN-γ)-induced galectin-9 expression in Kupffer cells plays an essential role in modulatingthe microenvironment of hepatitis-associated hepatocellular carcinoma (HCC). However, whether or not IFN-γ induces galectin-9 expression in HCC cells, its biological role and regulatory mechanism in HCC development and progression are poorly defined....

miR-221 silencing blocks hepatocellular carcinoma and promotes survival.

Patients with advanced hepatocellular carcinoma (HCC) face a dismal prognosis because of a lack of any effective therapies. To address this situation, we conducted a preclinical investigation of the therapeutic efficacy of oligonucleotides directed against the oncogenic microRNA miR-221, which has been implicated in HCC. Of 9 chemistries evaluated, we determined that a 2'-O-methyl phosphorothio...

miR-182-5p promotes hepatocellular carcinoma progression by repressing FOXO3a

BACKGROUND High frequency of recurrence is the major cause of the poor outcomes for patients with hepatocellular carcinoma (HCC). microRNA (miR)-182-5p emerged as a high-priority miRNA in HCC and was found to be related to HCC metastasis. Whether the expression of miR-182-5p in tumor tissue correlated with early recurrence in HCC patients underwent curative surgery was unknown. METHODS Real-t...
